Based in Weston, Massachusetts and operating as an OPKO Health company, ModeX Therapeutics was co-founded by Elias Zerhouni and Gary Nabel. Its MSTAR platform assembles antibodies addressing up to six distinct biological pathways in one molecule, retaining the antibody constant region for immune engagement, half-life, safety and manufacturability, while a simplified gene expression design also opens the door to gene-based antibody delivery. A second platform builds vaccines on naturally self-assembling ferritin, presenting 24 copies of a chosen antigen in a particle sized to reach immune cells in the lymph nodes. Clinical work is already under way: MDX2001, a tetraspecific T cell engager-expander, is in a Phase 1/2a first-in-human trial in advanced solid tumours, and MDX2004, a trispecific CD3xCD28/4-1BB antibody-fusion protein, has entered first-in-human dosing in advanced malignancies.
