Targeted protein degraders are advancing rapidly through the clinic, but measuring target degradation directly in patients remains a major bottleneck. In this webinar, Biodesix will present the development of a novel pharmacodynamic biomarker strategy to quantify degradation of a novel kinesin in peripheral blood mononuclear cells.

We'll walk through how discovery proteomics identified measurable novel kinesin peptides and confirmed selective degradation across cell models, how key pre-analytical variables were optimized to support reliable detection in clinical samples, and how the team built a targeted mass spectrometry assay sensitive enough to detect degradation at the low-attomole level.

Attendees will leave with a practical framework for building clinically viable pharmacodynamic biomarkers — from early discovery through a validated, deployment-ready assay.

Highlights:

• Discovery proteomics identified novel kinesin peptides that enable direct measurement of target abundance in PBMCs and demonstrated selective degradation in degrader-treated cell models.

• Pre-analytical studies evaluated blood collection, PBMC isolation, and sample handling variables influencing novel kinesin detection and assay performance.

• A targeted PRM assay achieved a 50 amol lower limit of quantification and reproducible performance across donor matrices, supporting pharmacodynamic assessment in clinical development.
